چکیده

We present here a LC-MS and GC-MS based analytical platform for the comprehensive analysis of cellular metabolites, including water-soluble metabolites, and waterinsoluble fatty acids, and phospholipids. The entire workflow consists of metabolic extraction, LC-MS and GC-MS runs, data analysis and interpretation. Standard operation procedures have been developed for the metabolite extraction from cell culture, tissue and serum/ plasma, which involve liquid extraction using appropriate extraction solvents. Metabolites were analyzed using multiple analytical methods on multiple dedicated instruments. Cationic water-soluble metabolites were analyzed on a triple quadrupole instrument using hydrophilic interaction chromatography [1]. Anionic water-soluble metabolites were analyzed using high resolution ‘Exactive’ Orbitrap mass spectrometer coupled to reversed phase ion pairing chromatography [2]. Fatty acids and phospholipids were analyzed using Agilent Q-TOF instrument coupled to reversed phase chromatography [3]. Data analyses were performed using MAVEN program which converts the raw data into a validated table of metabolite-specific signals [4]. Examples will be provided to demonstrate our capability to analyze a broad range of metabolites from real biological samples, as well as to probe the metabolic fluxes using stable isotope tracers.

Metabolomics, a Powerful Tool for Agricultural Research

Metabolomics, which is based mainly on nuclear magnetic resonance (NMR), gas-chromatography (GC) or liquid-chromatography (LC) coupled to mass spectrometry (MS) analytical technologies to systematically acquire the qualitative and quantitative information of low-molecular-mass endogenous metabolites, provides a direct snapshot of the physiological condition in biological samples. As complements...

Metabolic Profiling of Plasma from Benign and Malignant Pulmonary Nodules Patients Using Mass Spectrometry-Based Metabolomics

Solitary pulmonary nodule (SPN or coin lesion) is a mass in the lung and can be commonly found in chest X-rays or computerized tomography (CT) scans. However, despite the advancement of imaging technologies, it is still difficult to distinguish malignant cancer from benign SPNs. Here we investigated the metabolic profiling of patients with benign and malignant pulmonary nodules. A combination o...

MUSCLE: automated multi-objective evolutionary optimization of targeted LC-MS/MS analysis

Developing liquid chromatography tandem mass spectrometry (LC-MS/MS) analyses of (bio)chemicals is both time consuming and challenging, largely because of the large number of LC and MS instrument parameters that need to be optimized. This bottleneck significantly impedes our ability to establish new (bio)analytical methods in fields such as pharmacology, metabolomics and pesticide research. We ...
